酵母培养基配制Yeast media preparation
【液体培养基】 8g粉状培养基Cenvathys目录号CVAA013系列选择培养基加950 ml 水,常规高压灭菌后再加50ml 40%过滤除菌的无菌葡萄糖。(备注: YPD、YPDA和YPD plus这三种系列不需要再加糖,直接按瓶子上标签指示加水配制即可)。配制液体培养基不需要调pH,因为酵母偏好酸性条件生长。如果为了实验方便,葡萄糖也可按2%的量以粉末的形式加进去高压灭菌(见下面标准配制法注解)。
【固体培养基】8g粉状培养基(Cenvathys目录号CVAA013)加950 ml, 用NaOH调pH至大致范围5.8~6.5, 然后按20g/L加琼脂粉, 高压灭菌后再加预温的50ml 40%无菌葡萄糖, 摇匀后倾倒平皿(备注:如不调pH, 琼脂在酸性条件下会产生崩解)。如果为了实验方便,葡萄糖也可按2%的量以粉末的形式加进去高压灭菌。
【标准配制法】标准流程要求葡萄糖过滤除菌,因为葡萄糖与培养基中的氨基酸在高温高压下发生化学反应。但有些实验者为方便起见,将固体葡萄糖按2% 的量(按每升20克)直接加到培养基后一起高压灭菌,虽然酵母也可以生长,但一般不推荐这种将葡萄糖高压灭菌的方法。如果实在要图省事想用这种方法,那么一定要控制好高压时间在18-20分钟结束,待压力下降和温度降到安全范围后应尽快取出。否则液体颜色会变成黄色而对酵母特性发生不可预知的影响。但半乳糖和棉子糖不能高压灭菌,只能用过滤除菌制备,建议制备高浓度40%的储存液,按计算好的体积加到高压灭菌的培养基使得终浓度为2%。40%无菌棉子糖和半乳糖可以通过当地经销商从Cenvathys公司订购。
【说明】用于组成型表达(如酵母双杂交、酵母单杂交)以及其它采用组成型启动子的实验一般都加葡萄糖;诱导型表达如采用GAL1启动子载体的表达一般都加半乳糖和棉子糖。葡萄糖可以勉强高压灭菌,但半乳糖和棉子糖是绝对不能高压灭菌的, 应采用无菌过滤法先制备40%的无菌储备液,待不含糖的培养基高压灭菌后按最终应用浓度为2% 的量计算需要加入的无菌液体糖储存液:CVAA013-X系列酵母选择营养缺陷培养基Yeast selective media or selective minus media
